چکیده

The events responsible for cell injury after a tissue stimulation are only incompletely understood. The purpose of this study was to examine mechanisms of cell injury in two tissues, rat mesentery and cremaster muscle, after tissue stimulation with N-formylmethionyl-leucyl-phenylalanine (FMLP) and platelet-activating factor (PAF). The response was studied in the same animal in random order using normal and leukopenic rats. The tissues were exteriorized after pentobarbital anesthesia. Five to six vascularized areas were chosen in each tissue, and cell injury and hydroperoxide production were assessed visually by continuous superfusion with 1 μM propidium iodide and 5 μM dichlorofluorescin diacetate (DCFH), respectively. FMLP (1 × 10-8 M) and then PAF (1 × 10-8 M) were added to the superfusate, and measurements were made at several time points. The second tissue was then examined using the same protocol. In the cremaster, there was little hydroperoxide production, and the tissue injury was eliminated after leukopenia. Leukopenia had no effect on tissue injury in the mesentery. Although hydroperoxide production was observed, there was no correlation between it and the tissue injury. The level of preactivation showed no correlation with either tissue injury or hydroperoxide production. In light of these results, mast cell degranulation may be an important mechanism of tissue injury in the mesentery.

Establishing a new animal model for muscle regeneration studies

Skeletal muscle injuries are one of the most common problems in the worldwide which impose a substantial financial burden to the health care system.  Accordingly, it widely accepted that muscle regeneration is a promising approach that can be used to treat muscle injury patients. However, the underlying mechanisms of muscle regeneration have yet to be elucidated. The muscle structure and muscle...

Injury to skeletal muscle of mice following acute and sub-acute pregabalin exposure

Objective(s): Pregabalin (PGB) is a new antiepileptic drug that has received FDA approval for patient who suffers from central neuropathic pain, partial seizures, generalized anxiety disorder, fibromyalgia and sleep disorders. This study was undertaken to evaluate the possible adverse effects of PGB on the muscular system of mice. Materials and Methods: To evaluate the effect of PGB on skeletal...

Effects of velnacrine maleate in the leukocyte-endothelial cell interactions in rat cremaster microcirculatory network.

The expression of acetylcholinesterase in proinflammatory cells has supported the hypothesis that this protein plays a role in intercellular adhesion. Previous results of our group show that velnacrine, an acetylcholinesterase inhibitor, increases the number of adherent leukocytes in post-capillary venules of Wistar rats' mesentery muscle. This works intends to evaluate the local application of...

The effect of bone marrow mesenchymal stem cells on recovery of skeletal muscle after neurotization surgery in rat

Objective(s): When the nerve is injured near its entrance to the muscle belly, we cannot perform conventional methods. One useful method in such a situation is neurotization surgery. In this study, Bone marrow mesenchymal stem cells (BMSCs) implanted into the paralyzed muscle after neurotization surgery. These cells can stimulate axon growth and motor endplate formation, also prevent muscle atr...

Microvascular blood flow: evidence indicating a cubic dependence on arteriolar diameter.

The primary objectives of this study were 1) to determine the functional relationship between microvascular blood flow (Q) and arteriolar internal diameter (D) and 2) to determine whether this relationship conformed to a theoretical optimality prediction--that blood flow is proportional to the cube of the diameter (Q = kD3). Paired blood velocity and arteriolar diameter measurements in the crem...
